Basic Qualifications
Demonstrated expertise with standard aseptic cell culture technique, including experience culturing various primary cells. Experience with hepatocytes primary cell culture is highly appreciated. Experience with hepatocytes and/or liver non-parenchymal cells isolation is a plus.
Broad experience with cell-based functional assays, including transfection, cytotoxicity, cytokine release, and proliferation assays. Experience with co-culture in-vitro or MPS is a plus.
Proficiency across cellular, biochemical, and molecular techniques including protein detection (ELISA, MSD, western blot), gene profiling (RNA/DNA extraction, qPCR) and cell imaging. Experience with high content microscopy is a plus.
Broad scientific knowledge in liver biology and non-viral delivery and molecular biology.
Self-motivated with strong time management and organizational skills
Flexibility to rapidly changing priorities and deadlines
Experience in preparing high quality documents including technical reports, presentations, SOPs, and experimental protocols.
Highly collaborative with demonstrated ability to work in a matrixed team environment.
Excellent oral and written communication skills
Key Responsibilities:
Maintain cultures of immortalized tumor cell lines, hepatic primary cell types (e.g. hepatocytes, LSEC and KC).
Perform hepatocytes mouse isolation from key disease models.
Optimize, design, execute, and interpret in vitro cell-based assays to characterize diverse mechanisms of gene therapies (e.g. ELISA, MSD, high content imaging)
Perform fit-for-purpose in vitro and ex vivo assays to assess phenotypic and functional read-outs to assess drug candidate toxicity and efficacy.
Prepare experimental protocols, analyze experimental data, and present study results in team and department meetings.
Document experimental methods and findings in electronic laboratory notebook
POSITION OBJECTIVES
Conduct and interpret cell-based in vitro assays to characterize mechanism of action related to non-viral gene therapy with focus on the hepatic environment:
Immunomodulation
Cargo cell localization
Support non-viral (LNP) oligo therapy screenings e.g. endosomal scape assessment
